Background
BTN1A1 (also known as butyrophilin subfamily 1 member A1) is a 55 kDa type I transmembrane glycoprotein belonging to the immunoglobulin superfamily. Its structure includes extracellular IgV/IgC domains, a transmembrane region, and a cytoplasmic tail containing a B30.2 domain. Functionally, the B30.2 domain binds to xanthine oxidoreductase (XOR) to regulate milk fat globule secretion, while also serving as an intracellular phosphoantigen sensor to activate γδ T cells. Additionally, BTN1A1 inhibits CD4+ T cell proliferation, reduces the secretion of cytokines such as IL-2 and IFN-γ, and demonstrates protective effects in experimental autoimmune encephalomyelitis (EAE) models. Given its structural similarity to the B7 family and its involvement in immune regulation, BTN1A1 represents a potential novel co-stimulatory/inhibitory molecule and a promising therapeutic target for autoimmune and immune-related diseases.
