Background
BSCL2/Seipin is an endoplasmic-reticulum-anchored multi-pass transmembrane protein that drives proper lipid-droplet biogenesis by catalyzing the clustering and directed budding of nascent neutral-lipid lenses within the ER membrane, thereby acting as a molecular scaffold that recruits and concentrates diacylglycerol acyltransferase, phosphatidic-acid-metabolizing enzymes and lipin-1 to nascent droplet formation sites, while missense mutations that disrupt its N-glycosylation motif trigger ER stress, ubiquitin–proteasome degradation and conformational seipinopathy leading to congenital generalized lipodystrophy type 2 or dominant motor-neuron degenerations.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human spinal cord. Anti-BSCL2/Seipin antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
