Background

BMP6 is a secreted signaling molecule belonging to the transforming growth factor-β (TGF-β) superfamily. Synthesized as a precursor protein, BMP6 undergoes proteolytic processing to form a disulfide-linked homodimer that exerts its biological functions in a paracrine or endocrine manner. In terms of its classical functions, BMP6 is a potent inducer of cartilage and bone formation, involved in osteoblast differentiation and skeletal development, and also regulates various physiological processes including adipogenesis and ovulation. However, in recent years, the role of this protein in systemic iron metabolism regulation has garnered significant attention—as a ligand of the bone morphogenetic protein (BMP) signaling pathway, BMP6 binds to the co-receptor hemojuvelin (HJV) and the type I receptor ALK3 (BMPR1A), activating the SMAD signaling cascade, thereby upregulating hepatic hepcidin expression to precisely regulate systemic iron homeostasis. In addition, BMP6 can modulate vascular endothelial growth factor (VEGF) signaling through the TAZ-Hippo non-canonical pathway and participates in a wide range of processes including inflammatory responses, immune reactions, and cellular responses to mechanical stimuli. Given its critical nodal role in both bone formation and iron metabolism, dysregulation of BMP6 function is closely associated with iron overload disorders such as hereditary hemochromatosis, as well as with the progression of various cancers.
