Background

Bone morphogenetic protein 2 (BMP-2) is a multifunctional growth factor belonging to the transforming growth factor-β (TGF-β) superfamily. The osteoinductive activity of bone-derived proteins was first described by Marshall Urist in 1965. As one of the most potent osteoinductive factors known, BMP-2 initiates the differentiation of mesenchymal stem cells into osteoblasts and chondrocytes, playing essential roles in embryonic skeletal development, bone remodeling, and fracture healing.

In the biomedical industry, rhBMP-2 received FDA approval in 2002 and is commercially available as the Infuse™ Bone Graft system, delivered via an absorbable collagen sponge carrier. Industrial applications are predominantly focused on spinal fusion surgeries, where rhBMP-2 has demonstrated fusion rates comparable to or higher than autologous bone grafts. Additional clinical indications include open tibial fractures, maxillofacial reconstruction, sinus augmentation, and treatment of femoral head necrosis. However, clinical use is constrained by dose-dependent complications including inflammation, ectopic ossification, osteolysis, and concerns regarding cost and safety.

Determined by its ability to induce alkaline phosphatase production by ATDC-5 cells. The expected EC50 for this effect is 100-144 ng/ml.

Measured by its ability to induce BMP-2 responsive firefly luciferase reporter activity in HEK293T human embryonic kidney cells. The expected EC50 for this effect is 0.38-3.3 ng/ml.
