Background
Vascular endothelial growth factor receptor 1 (VEGFR-1), also known as Fms-like tyrosine kinase-1 (FLT1), is a key member of the VEGF receptor family. Structurally, it comprises seven extracellular immunoglobulin-like domains, a transmembrane region, and an intracellular tyrosine kinase domain, and exists in two isoforms: membrane-bound VEGFR-1 and a soluble splice variant (sFLT1). VEGFR-1 binds VEGF-A, VEGF-B, and PlGF, and regulates not only angiogenesis and endothelial function but also inflammation, macrophage activation, and neuronal survival. sFLT1 acts as a decoy receptor that negatively modulates VEGF signaling, and its dysregulation is closely associated with preeclampsia and diabetic nephropathy. Membrane-bound VEGFR-1 plays complex roles in tumor angiogenesis, neuronal injury in Alzheimer's disease, and cardiovascular disorders, making it an important target for gene therapy and anti-angiogenic therapeutics.
