Background

Nucleolin (NCL) is a crucial multifunctional nucleolar protein in eukaryotic cells, possessing nucleo-cytoplasmic shuttling capability. Its three distinctive structural domains enable it to induce chromatin decondensation by binding to histone H1 and to specifically recognize RNA sequences (with higher affinity for 5'-UUAGGG-3' repeats than their DNA counterparts). This protein not only participates in fundamental biological processes such as ribosome biogenesis, pre-rRNA transcription and processing, and transcriptional elongation regulation, but is also extensively involved in cell proliferation, apoptosis regulation, and stress response. Under pathological conditions, through its roles in tumorigenesis and viral infection, nucleolin has emerged as a significant potential target for anti-tumor and anti-viral therapies.
