Background
ITGAV (integrin subunit alpha V) and ITGB8 (integrin subunit beta 8) non-covalently pair to form the αvβ8 heterodimer, the only integrin in which β8 exclusively associates with αv. It is minimally expressed in normal tissues but upregulated in solid tumors and fibrosis. Functionally, αvβ8 binds latent TGF-β complexes and mediates local TGF-β activation, driving an immunosuppressive tumor microenvironment and fibrosis; it also regulates microglial development and serves as a viral attachment receptor. Clinically, the αvβ8-blocking antibody CRB-601 has entered phase I/II trials in solid tumors, and the oral αvβ8/αvβ1 inhibitor PLN-101095 combined with immune checkpoint inhibition shows synergistic antitumor effects. ITGB8 is overexpressed in ovarian cancer and promotes invasion via PI3K-AKT-NF-κB, serving as a potential prognostic biomarker. These advances support αvβ8-targeted therapy and diagnostics.
