Background

IL-2 Rγ/CD132 is a transmembrane protein belonging to the type I cytokine receptor family, characterized by a highly conserved extracellular WSXWS motif, a transmembrane region, and an intracellular domain containing Box1/Box2 motifs. It serves as the shared signaling chain for six cytokines: IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21. Through its intracellular domain, it couples with signaling molecules such as JAK3 and STAT5, playing an indispensable core role in T/NK cell development, lymphocyte homeostasis, and immune regulation. The clinical significance of this molecule is substantial—mutations in its gene cause X-linked severe combined immunodeficiency (X-SCID). Therapeutic strategies targeting this molecule have been translated in three key ways: gene therapy encoding γc (e.g., Strimvelis) has received clinical approval; CD132-targeting bispecific antibodies selectively activate the common receptor complex within the tumor microenvironment to achieve anti-tumor immunity while avoiding systemic toxicity; and engineered IL-2/antibody biased complexes modulate CD132-dependent signaling preferences to expand regulatory T cells for transplant tolerance and autoimmune disease treatment.
