Background
Interleukin-13 (IL-13) is a type I cytokine secreted by Th2 cells, mast cells, basophils, and other immune cells, belonging to the IL-4/IL-13 cytokine family. It exerts its biological functions through two distinct receptor complexes: the type I receptor (IL-4Rα/IL-13Rα1) primarily mediates signal transduction, whereas the type II receptor (IL-13Rα2) functions as a decoy receptor that negatively regulates signaling or serves as a tumor-specific therapeutic target. IL-13 is a central driver of type 2 immune responses, participating in IgE production, eosinophil recruitment, mucus secretion, and tissue fibrosis. Currently, IL-13-targeted biologics have achieved significant breakthroughs: selective IL-13 inhibitors such as tralokinumab and lebrikizumab have been approved for the treatment of moderate-to-severe atopic dermatitis; meanwhile, IL-13Rα2, which is highly expressed in glioblastoma, pancreatic cancer, and other solid tumors, has emerged as an important tumor-specific target for the development of CAR-T cell therapy, immunotoxins, and antibody-drug conjugates.
