Background

The interferon-α/β receptor alpha chain (IFNAR1, CD118) is a single-pass transmembrane protein that belongs to the class II cytokine receptor family and contains four fibronectin type-III domains; together with IFNAR2 it forms the type I interferon receptor. Serving as the high-affinity binding site for IFN-α/β, IFNAR1 can also transduce IFN-β signals independently and modulate—sometimes suppress—the Jak-STAT pathway, thereby acting as a critical on/off switch in innate antiviral immunity. Its expression level or mutation at Tyr466 directly affects STAT phosphorylation efficiency, linking IFNAR1 to chronic viral infections, autoimmune disorders and interferon therapy resistance, making it a key biomarker for assessing IFN efficacy and developing targeted immunomodulatory drugs.
