Background

Cynomolgus macaque (Macaca fascicularis) CD8A shares high sequence homology with human CD8α (approximately 95%), and its CD8+ T cells are highly conserved with humans in terms of immunophenotype, signal transduction, and function. CD8α forms an Ig-like domain dimer that binds to the α3 domain of MHC class I molecules through CDR-like loops, thereby enhancing the recognition affinity of TCR for antigens. Cynomolgus CD8+ T cells serve as a critical model for evaluating HIV/SIV vaccines, immune checkpoint inhibitors (such as anti-PD-1/PD-L1), and bispecific antibodies (such as B7-H4×CD3). The cynomolgus macaque model is utilized to predict the toxicity (such as cytokine release syndrome) and efficacy of immunotherapeutic drugs, wherein CD8+ T cell depletion experiments represent the gold standard for validating vaccine protective mechanisms and the effectiveness of immunotherapy targets.
