Flow cytometric analysis of Jurkat cells (Human T cell leukemia T lymphocyte). Cells were treated for 4 hours with 6 µM Camptothecin and labeled with Annexin V His Tag Protein (1 μg). Then cells were stained with anti-His Tag APC.Then cells were stained with 7-AAD. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Annexin V (also known as Annexin A5) is a calcium-dependent phospholipid-binding protein with a molecular weight of approximately 35 kDa. Its structure comprises four highly conserved repeat domains (each containing approximately 70 amino acids), with a core region harboring binding sites for both calcium ions and phosphatidylserine (PS). Functionally, Annexin V binds with high affinity to PS exposed on the outer leaflet of apoptotic cells, serving as the "gold standard" tool for apoptosis detection; meanwhile, it also exhibits multiple biological activities including anticoagulation, immune modulation, and tissue protection. In recent years, the clinical applications of Annexin V have expanded from simple apoptosis detection to targeted therapeutics—by leveraging its specific recognition of apoptotic cells, researchers have conjugated Annexin V with drugs, nanoparticles, or radionuclides for tumor-targeted drug delivery, neuroprotection following ischemic stroke, and cardiovascular disease treatment, demonstrating broad prospects in translational medicine.
