Flow cytometric analysis of Mouse CD366 (TIM-3) expression on mouse CD3, CD28 antibody treated C57BL/6 mouse splenocytes. C57BL/6 mouse splenocytes were either not stimulated (Left panel) or stimulated 3 days with anti-mouse CD3 and anti-mouse CD28 (Right panel) and then stained with Brilliant Violet 421™ Rat Anti-Mouse CD4 and SDT Biotin Rat Anti-Mouse CD366 (TIM-3) antibody at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
TIM-3 (CD366) is a trans-membrane immune checkpoint receptor in the Ig super-family whose extracellular domain binds phosphatidyl-serine, CEACAM-1 or galectin-9 ligands to trigger downstream signaling through its conserved cytoplasmic tyrosine motif, thereby attenuating T-cell, NK-cell and dendritic-cell activation while promoting regulatory T-cell function and myeloid tolerance, making its blockade a promising strategy for enhancing anti-tumor and anti-viral immunity across multiple cancers and chronic infections.
