Flow cytometric analysis of Mouse Siglec-E expression on BALB/c mouse splenocytes. BALB/c mouse splenocytes were stained with Pacific Blue™ Rat Anti-Mouse Ly-6G/Ly-6C antibody and either Biotin Rat IgG2b Isotype Control (Left panel) or SDT Biotin Rat Anti-Mouse Siglec-E Antibody (Right panel) at 5 μl/test followed by Sav-PE. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Siglec-E, the murine orthologue of human Siglec-9, is a sialic acid-binding immunoglobulin-like lectin expressed predominantly on myeloid cells (macrophages, neutrophils, dendritic cells) that functions as a critical immunosuppressive checkpoint by recruiting SHP-1/2 phosphatases upon engagement of sialoglycan ligands, thereby attenuating TLR4- and TRIF-dependent signaling, dampening NF-κB activation and pro-inflammatory cytokine production, and exerting neuroprotective and anti-atherogenic effects through inhibition of microglial activation and CD36-mediated oxidized LDL uptake.
