Flow cytometric analysis of Ly-6B.2 expression on fresh C57BL/6 mouse splenocytes. Fresh C57BL/6 mouse splenocytes were stained with Alexa Fluor® 647 Rat Anti-Mouse CD11b Antibody and either Biotin Rat IgG2a, k Isotype Control (left panel) or SDT Biotin Rat Anti-Mouse Ly-6B.2 Antibody at 2 μl/test followed by Sav-iFluor 488. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
Ly-6B.2 is an important glycosylphosphatidylinositol (GPI)-anchored protein belonging to the mouse Ly-6 antigen complex family, with a molecular weight of approximately 25–30 kDa, which reduces to about 15 kDa upon removal of N-linked glycosylation. This protein is predominantly highly expressed on the surface of neutrophils, inflammatory monocytes, and partially activated macrophages, and is abundantly present in tissues such as bone marrow, spleen, lung, and lymph nodes. The function of Ly-6B.2 is closely associated with inflammatory responses; its high expression on inflammatory cells makes it an important participant in inflammatory reactions. Studies have shown that Ly-6B-expressing macrophages represent a newly generated population of inflammatory macrophages and serve as a surrogate marker for macrophage turnover at inflammatory foci.
