Flow cytometric analysis of Mouse Ly-6A/E expression on C57BL/6 mouse splenocytes. C57BL/6 mouse splenocytes were stained with either Biotin Rat IgG2a, κ Isotype Control (Left panel) or SDT Biotin Rat Anti-Mouse Ly-6A/E Antibody (Right panel) at 5 μl/test followed by Sav-iFluor 488. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Ly-6A/E, also known as Sca-1 (stem cell antigen-1), is an 18-kDa glycosylphosphatidylinositol (GPI)-anchored cell surface protein belonging to the murine Ly-6 multigene family, predominantly expressed on multipotent hematopoietic stem cells in adult mouse bone marrow and fetal liver, and is widely used as a marker for isolating these stem cells; beyond its stem cell association, Ly-6A/E is upregulated on activated T and B lymphocytes and appears to participate in T-cell receptor-mediated activation, myoblast proliferation and differentiation, bone formation, and interferon-induced immune responses, with expression levels varying between mouse strains carrying different Ly-6 haplotypes.
