Background

IL-17A protein a is key cytokine produced by various immune cells such as Th17 cells. It plays a crucial role in the immune response by promoting inflammation. It can induce the expression of various pro-inflammatory molecules in different cell types, including chemokines that recruit neutrophils and other immune cells to the site of infection or injury. IL-17A is involved in the defense against certain pathogens, but its excessive production is also associated with the pathogenesis of several autoimmune and inflammatory diseases like psoriasis and rheumatoid arthritis. Targeting IL-17A has become an important therapeutic strategy in the treatment of these conditions.

Flow cytometric analysis of IL-17A expression in C57BL/6 mouse splenocytes polarized with Th17 Polarization Kit, Mouse (S0B090020). Th17 polarized C57BL/6 mouse splenocytes were harvested and restimulated for 5 hr with PMA and Ionomycin in the presence of Brefeldin A (Right Panel) or unstimulated (Left Panel).
The cells were harvested and fixed with 4% PFA and permeabilized with Intracellular Fixation & Permeabilization Buffer Set. The cells were then stained with SDT Biotin Rat Anti-Mouse IL-17A Antibody at 1.25μl/test followed by Sav-PE. Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
