Flow cytometric analysis of Mouse CD370 expression on C57BL/6 mouse splenocytes. C57BL/6 mouse splenocytes were stained with FITC Rat Anti-Mouse CD11c antibody and APC Rat Anti-Mouse CD8α either Biotin Rat IgG2b, κ Isotype Control (Left panel) or SDT Biotin Rat Anti- Mouse CD370 antibody (Right panel) at 5 μl/test followed by Sav-PE. Gated on CD8+ viable cells. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD370, also known as CLEC9A or DNGR-1, is a type II transmembrane glycoprotein expressed almost exclusively on human CD141⁺ (mouse CD8α⁺) dendritic cells and a minor subset of CD14⁺CD16⁻ monocytes; it functions as an endocytic pattern-recognition receptor that binds exposed filamentous actin on necrotic cell debris, triggering Syk–ITAM signaling that drives efficient cross-presentation of dead-cell antigens to cytotoxic T lymphocytes and thereby links necrosis sensing to initiation of adaptive immunity.
