Flow cytometric analysis of Mouse CD184 expression on C57BL/6 mouse splenocytes. C57BL/6 mouse splenocytes were stained with APC/Fire™ 750 Rat Anti-Mouse CD4 Antibody and either Biotin Rat IgG2b, κ Isotype Control (left panel) or SDT Biotin Rat Anti-Mouse CD184 Antibody (right panel) at 2 μl/test followed by Sav-iFluor 488. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

CD184, also known as CXCR4, is a seven-transmembrane G-protein-coupled chemokine receptor that—after binding its unique ligand CXCL12 (SDF-1)—triggers multiple signaling cascades (PI3K/AKT, JAK/STAT, ERK1/2, Hedgehog) governing immune cell trafficking, plasma-cell homing to bone-marrow niches, embryonic development, hematopoiesis, angiogenesis, and cancer stem-cell maintenance, while its overexpression or mutations underlie WHIM syndrome, HIV entry, tumor progression and metastasis, making it a key therapeutic target in oncology and immunology.
