Flow cytometric analysis of TOX expression in C57BL/6 mouse thymocytes. C57BL/6 mouse thymocytes were fixed and permeabilized with Foxp3 / Transcription Factor Staining Buffer Set. The cells were then stained with Pacific Blue™ Rat Anti- Mouse CD8 Antibody and either Biotin Rat IgG1, k Isotype Control (left panel) or SDT Biotin Rat Anti- Human/Mouse TOX Antibody (right panel) at 5 μl/test followed by Sav-PE. Total viable cells, as determined by Fixable Viability Dye 545 (S0B88802), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
TOX protein is a transcription factor that plays a crucial role in the immune system. It is involved in the regulation of T cell differentiation and function, particularly in the development and maintenance of regulatory T cells (Tregs) which are essential for maintaining immune tolerance and preventing autoimmunity. Additionally, TOX has been implicated in the response of T cells to chronic antigen stimulation, such as in the context of chronic infections or cancer, where it may contribute to T cell exhaustion. Its expression and activity can influence the balance between effector and regulatory T cell populations, thereby having a significant impact on the overall immune response.
