Background

XCR1 is a conserved, 7-transmembrane G-protein-coupled receptor selectively expressed on CD8α⁺ and CD103⁺ classical dendritic cells; its exclusive ligand XCL1 is secreted by activated CD8⁺ T and NK cells, forming a chemotactic feedback loop that recruits antigen-loaded DCs into lymphoid tissue and optimizes cross-presentation, Th1 cytotoxicity and antiviral/antitumor immunity, making the XCR1–XCL1 axis an emerging target for cancer immunotherapy and vaccine adjuvant design.

Flow cytometric analysis of human XCR1 expression on Human PBMC (human peripheral blood mononuclear cells). Human PBMC were stained with Brilliant Violet 421™ Mouse Anti-Human CD141 Antibody and either Biotin Rat IgG2a, k Isotype Control (left panel) or SDT Biotin Rat Anti-Human XCR1 Antibody (right panel) at 5 μl/test followed by Sav-iFluor 488. Total viable cells, as determined by Fixable Viability Dye 662 (S0D0016), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
