Background

Siglec-8 is a CD33-related cell-surface lectin selectively expressed on human eosinophils and, at lower density, on mast cells and basophils; it carries an immunoreceptor tyrosine-based inhibitory motif (ITIM) and an ITIM-like motif that, upon engagement by sialylated glycan ligands, recruit SHP-1/2 phosphatases to deliver a potent inhibitory signal that suppresses FcεRI- or cytokine-driven activation, survival and mediator release, thereby serving as a key off-switch for allergic inflammation and making it an attractive therapeutic target for eosinophilic asthma, chronic urticaria and other IgE-mediated diseases.

Flow cytometric analysis of human Siglec-8 expression on human peripheral blood cells. Human peripheral blood cells were stained with PE Mouse Anti-Human CD193 Antibody and either Biotin Mouse IgG1, κ Isotype Control (left panel) or SDT Biotin Mouse Anti-Human Siglec-8 Antibody (right panel) at 5 μl/test followed by Sav-APC. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
