Background

CD61, also known as integrin beta-3 (ITGB3) or glycoprotein IIIa (gpIIIa), is a transmembrane protein encoded by the ITGB3 gene in humans. It is predominantly expressed on the surface of platelets and megakaryocytes. CD61 forms a crucial complex with CD41 (alphaIIb) to create the heterodimeric receptor gpIIb/IIIa, which is essential for platelet aggregation and thrombus formation. This receptor binds to fibrinogen, von Willebrand factor, and other extracellular matrix proteins, facilitating platelet adhesion and aggregation. In addition to its role in hemostasis, CD61 is also involved in various cellular processes, including cell adhesion, migration, and proliferation. It has been implicated in tumor progression, where it can influence tumor metabolism, immune microenvironment, and epithelial-to-mesenchymal transition.

Flow cytometric analysis of CD61 expression on fresh human peripheral blood cells. Fresh human peripheral blood cells were stained with APC Mouse Anti-Human CD41 Antibody and either Biotin Mouse IgG1, k Isotype Control (left panel) or SDT Biotin Mouse Anti-Human CD61 Antibody (right panel) at 5 μl/test followed by Sav-iFluor 488. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
