Flow cytometric analysis of human CD126 expression on human peripheral blood lymphocytes. Human peripheral blood lymphocytes were stained with Brilliant Violet 421™ Mouse Anti-Human CD3 Antibody and either Biotin IgG1, κ Isotype Control (left panel) or SDT Biotin Mouse Anti-Human CD126 Antibody (right panel) at 5μl/test followed by Sav-PE. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
CD126, also known as the interleukin-6 receptor alpha chain (IL-6Rα), is a type I cytokine receptor that plays a crucial role in regulating cell growth and differentiation in various tissues. It is a subunit of the IL-6 receptor complex, which also includes the signal-transducing subunit glycoprotein 130 (gp130 or CD130). CD126 is primarily expressed on B cells, epithelial cells, and hepatocytes. The binding of IL-6 to CD126 forms a complex that, in association with CD130, initiates signaling pathways involved in immune responses and acute phase reactions. Dysregulated production of IL-6 and CD126 is implicated in the pathogenesis of several diseases, including multiple myeloma, autoimmune diseases, and certain cancers. Additionally, CD126 is being explored as a potential therapeutic target for CAR-T cell therapy due to its expression in various tumors.
