Background

Beta Tubulin is a highly conserved and essential globular protein in the eukaryotic cytoskeleton, serving as one of the core structural components that constitute microtubules—hollow tubular polymers—typically in the form of heterodimers alternately arranged with alpha-tubulin. As a member of the GTPase family, beta-tubulin drives microtubule dynamic instability—the stochastic switching between growth (polymerization) and shrinkage (depolymerization) at microtubule ends—through GTP binding and hydrolysis, a property that is critical for various life activities including spindle formation during cell division, directed intracellular transport of vesicles and organelles (via molecular motors such as dynein and kinesin), and maintenance of cell morphology and polarity. In mammals, beta-tubulin is encoded by multiple genes with tissue-specific expression patterns (such as TUBB, TUBB3, etc.), and the subtle sequence differences among these isotypes, together with post-translational modifications (e.g., phosphorylation, acetylation, polyglutamylation), constitute the "tubulin code" that finely regulates microtubule physical stability and interactions with other proteins. Clinically, beta-tubulin is a direct molecular target for various anti-tumor drugs—including taxanes such as paclitaxel and docetaxel, which bind to and stabilize microtubules to prevent depolymerization, and vinca alkaloids, which inhibit microtubule polymerization—while mutations or overexpression of beta-tubulin genes (particularly TUBB3) are frequently associated with acquired resistance to these chemotherapeutic agents in tumors, making it both an important pharmacological target and a key predictive biomarker for therapeutic efficacy in cancer treatment.
