Background
Beta III tubulin (TUBB3) is a subtype of the beta-tubulin family, encoded by the TUBB3 gene, and plays a critical role in the development and function of the mammalian nervous system. As a neuron-specific cytoskeletal protein, it is highly expressed in the cell bodies, axons, and growth cones of embryonic and early postnatal neurons. Beyond participating in microtubule polymerization and stabilization, as well as maintaining neuronal morphology and polarity, its highly dynamic properties facilitate axon growth, guidance, and synaptic plasticity. Due to its stable and sustained expression in both immature and mature neurons, Beta III tubulin is widely used as a specific marker of the neuronal lineage in studies of neural development, neurotoxicity testing, and tumor classification (e.g., distinguishing neuroblastoma and neuroendocrine tumors).

Immunocytochemistry
ICC shows positive staining in SH-SY5Y cells. Anti-Beta III tubulin (Alexa Fluor® 488 Conjugate) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C.The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
Immunofluorescence
IF shows positive staining in paraffin-embedded human cerebellum. Anti-Beta III tubulin (Alexa Fluor® 488 Conjugate) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
