Background
Beta I tubulin, the most neuron-enriched β-tubulin isoform encoded by TUBB1, is a ~50 kDa globular protein that heterodimerizes with α-tubulin to assemble neuron-specific microtubule arrays, stabilizing axonal and dendritic cytoskeletons through GTP-dependent head-to-tail polymerization and interaction with microtubule-associated proteins; its unique C-terminal tail undergoes reversible glutamylation and detyrosination to regulate motor-protein trafficking, synaptic plasticity and axonal transport, while missense mutations (e.g., V180A, R215W) disrupt microtubule dynamics and cause a spectrum of neurodevelopmental disorders termed “tubulinopathies” characterized by simplified gyral pattern, microcephaly and intellectual disability, making Beta I tubulin a critical determinant of neuronal morphology and human brain development.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
