Background
BAP1 (BRCA1-associated protein 1) is an important tumor suppressor gene whose encoded protein belongs to the ubiquitin carboxyl-terminal hydrolase family. Its main function is to act as a deubiquitinating enzyme that removes ubiquitin molecules from substrates such as histone H2A, thereby participating in the regulation of gene expression, the cell cycle, DNA damage repair, and cell death, among other key cellular processes. BAP1 typically forms a multi-protein complex (PR-DUB) with proteins such as ASXL1/2, and through deubiquitination of H2A, it finely regulates chromatin status and transcriptional activity, thereby exerting its tumor-suppressive effects. When BAP1 acquires germline mutations or loss of function, it significantly increases an individual's risk of developing various tumors, including uveal melanoma, malignant mesothelioma, and clear cell renal cell carcinoma. This phenomenon is known as "BAP1 tumor predisposition syndrome."
