Background
BAFF (B-cell activating factor) and APRIL (a proliferation-inducing ligand) are members of the tumor necrosis factor (TNF) superfamily. They typically exist as homotrimers but can also form heterotrimers. Secreted by activated myeloid cells, they exert immunomodulatory effects by regulating B-cell differentiation, proliferation, survival, and effector functions. Clinically, dysregulated expression of the BAFF/APRIL system is closely associated with autoimmune diseases, particularly with significantly elevated levels observed in patients with systemic lupus erythematosus (SLE). The detection of circulating heterotrimers in the serum of patients with systemic immune rheumatic diseases suggests that these heterotrimers may represent a novel therapeutic target for rheumatic and autoimmune disorders.
