Protocol
Add PBMCs to 24-well/48-well/6-well cell culture plates or dishes according to individual demands. Adjust the cell density to 5e4-2e5/ml, cell culture medium is RPMI-1640 supplemented with 10% inactivated human serum, 55 μM β-mercaptoethanol , 100ng/ml IL-2, 100ng/ml IL-10, 100ng/ml IL-21 and feeder cells. After 5 days, observe and collect the cells for flow cytometry analysis or other applications.
Negative control: PBMC cultured in RPMI-1640, 55 μM β-mercaptoethanol and 10% inactivated human serum for 5 days.
