Background
ATGL (adipose triglyceride lipase, also known as PNPLA2) is the rate-limiting enzyme that initiates the lipolysis process in animal cells, primarily responsible for catalyzing the hydrolysis of triglycerides into diglycerides and free fatty acids—this initial step represents the most critical regulatory node in the entire lipolytic cascade. When energy demand increases, ATGL activity is finely regulated: its intrinsic activity is significantly enhanced by binding to CGI-58 protein; however, when cells are in a high-energy state, G0S2 protein binds to ATGL and inhibits its function, while phosphorylation modifications also play an important role in modulating its activity. ATGL not only governs energy metabolism in adipose tissue but also regulates lipid homeostasis in various organs including the heart, liver, and skeletal muscle. Dysfunction of ATGL is closely associated with multiple metabolic diseases such as obesity, insulin resistance, fatty liver disease, and myocardial lipotoxicity, making it an important potential therapeutic target for metabolic syndrome and cardiovascular diseases.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded mouse brown fat. Anti-ATGL antibody was used at 1/100 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Immunofluorescence
IF shows positive staining in paraffin-embedded mouse brown fat. Anti-ATGL antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
