Background

Atg3 is an E2-like enzyme essential for autophagosome biogenesis: its C-terminal catalytic cysteine forms a thioester bond with the C-terminal glycine of Atg8-family proteins (LC3/GABARAPs) in an ATP-dependent manner, and subsequently, with the E3-like Atg12–Atg5-Atg16L1 complex, it covalently conjugates Atg8 to phosphatidylethanolamine on the expanding phagophore membrane, thereby driving membrane curvature, expansion, and closure into the double-membrane autophagosome that sequesters cytoplasmic cargo for lysosomal degradation.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-Atg3 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- Atg3 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
