Background

ATF1 (activating transcription factor 1) is a 271-amino-acid basic leucine-zipper (bZIP) protein that forms homo- or heterodimers with c-Jun, CREB or other bZIP partners to bind the consensus cAMP response element 5′-TGACGTCA-3′, thereby functioning as a stimulus-inducible transcriptional activator in cAMP, MAPK, calcium and stress-signaling cascades that govern proliferation, survival, melanocyte differentiation, synaptic plasticity and memory; it is subject to phosphorylation-driven control (notably by PKA at Ser63 and by p38/MSK1 at Ser63/121), acetylation and ubiquitination, is frequently fused to either EWSR1 or FUS in Ewing sarcoma and angiomatoid fibrous histiocytoma through t(12;22) or t(12;16) chromosomal translocations that convert it into an aberrant transcriptional activator, and its locus at 12q13 is co-amplified with CDK4 in a variety of sarcomas, underscoring its dual role as both a tightly regulated physiological transcription factor and an oncogenic driver.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-ATF1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
