Background

ARNT2 is a transcription factor belonging to the basic helix-loop-helix/Per-Arnt-Sim (bHLH-PAS) superfamily, which contains a bHLH domain and two PAS domains and is localized in the nucleus. This protein serves as a critical dimerization partner, forming heterodimers with various bHLH-PAS family sensor proteins such as hypoxia-inducible factor 1α, single-minded homologs SIM1 and SIM2, and neuronal PAS domain protein 4 (NPAS4), thereby recognizing and binding to specific regulatory DNA sequences (e.g., hypoxia response elements and xenobiotic response elements) in the promoters or enhancers of target genes to regulate gene expression. Functionally, ARNT2 plays a key role primarily by mediating hypoxia signaling pathways and participating in the development of the central nervous system, particularly the hypothalamic-pituitary axis. Unlike the ubiquitously expressed ARNT1, ARNT2 exhibits tissue-specific expression and is predominantly restricted to the brain and kidneys in adult mice. Loss-of-function mutations in this gene cause the rare autosomal recessive disorder Webb-Dattani syndrome, characterized by hypothalamic-pituitary axis hypoplasia, global developmental delay, and visual and renal abnormalities. Furthermore, the functions of ARNT2 are increasingly recognized to be associated with various pathological processes, including inflammation, cardiovascular diseases, metabolic disorders, and multiple cancers.
