Background

ARIH1 (Ariadne RBR E3 ubiquitin-protein ligase 1) is a 557-amino-acid RING-between-RING (RBR) E3 ligase that ubiquitinates multiple substrates—most notably DNA-PKcs, whose proteasomal degradation activates the cGAS-STING pathway to enhance anti-tumor immunity, while in colorectal cancer it instead promotes K63-linked ubiquitination of PHB1 to drive mitochondrial respiration and tumor progression—thereby acting as a context-dependent regulator of proliferation, DNA repair, epithelial-mesenchymal transition and immune evasion through its dynamic interactions with E2 enzymes and cullin-RING ligase complexes.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- ARIH1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

ICC shows positive staining in NIH/3T3 cells. Anti- ARIH1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
