Background

ARF6 (ADP-ribosylation factor 6) is a small GTPase belonging to the ARF family that functions as a critical molecular switch regulating plasma membrane dynamics, endosomal trafficking, and actin cytoskeleton remodeling by cycling between active GTP-bound and inactive GDP-bound states. Unlike other ARF isoforms primarily associated with Golgi transport, ARF6 uniquely localizes to the plasma membrane and endosomes, where it orchestrates clathrin-independent endocytosis, recycling of membrane proteins such as integrins and MHC class I molecules, and the formation of membrane ruffles and lamellipodia through downstream effectors like JIP3/4, POR1, and phospholipase D. Its activity is tightly controlled by guanine nucleotide exchange factors (GEFs) such as EFA6 and cytohesins, as well as GTPase-activating proteins (GAPs) including ACAPs and GITs, ensuring precise spatiotemporal regulation of cellular processes vital for cell migration, adhesion, phagocytosis, and pathogen entry, while its dysregulation has been implicated in cancer metastasis, neurological disorders, and infectious diseases due to aberrant membrane trafficking and cytoskeletal reorganization.
