Flow cytometric analysis of Mouse Ly-6C expression on C57BL/6 mouse bone marrow. C57BL/6 mouse bone marrow was stained with Brilliant Violet 421™Rat Anti-Mouse CD11b Antibody and either APC Rat IgG2c, κ Isotype Control (Left panel) or SDT APC Rat Anti-Mouse Ly-6C Antibody (Right panel) at 1μl/test. Total viable cells, as determined by Fixable Viability Dye 515 (S0D0013), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Ly-6C is a glycosylphosphatidylinositol-anchored cell-surface protein belonging to the Ly-6/uPAR/α-neurotoxin superfamily, selectively expressed on murine monocyte and T-cell subsets where it serves as a dynamic lineage and activation marker—its high level on pro-inflammatory “classical” monocytes (CD11b⁺Ly-6C^hi) enables their recruitment via CCR2 to inflamed tissues, whereas Ly-6C^lo monocytes patrol vasculature and replenish resident macrophages, and transient up-regulation on CD8⁺ T cells correlates with effector differentiation; structurally it forms a three-finger α-helical bundle that can laterally associate with lipid rafts and signaling partners such as T-cell receptor or integrin complexes to modulate calcium flux, cytokine release, and adhesion, thereby critically steering innate–adaptive immune crosstalk in infections, autoimmunity, and cardiovascular disease.
