Flow cytometric analysis of human CD197 expression on Human PBMC (human peripheral blood mononuclear cells). Human PBMC were stained with Brilliant Violet 421™ Mouse Anti-Human CD4 Antibody and either Allophycocyanin Mouse IgG2a, κ Isotype Control (left panel) or SDT Allophycocyanin Mouse Anti-Human CD197 Antibody (right panel) at 5 μl/test. Total viable cells, as determined by Fixable Viability Dye 515 (S0D0013), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD197, also known as CCR7 (C-C chemokine receptor type 7), is an important G protein-coupled receptor primarily expressed on mature dendritic cells, naïve T cells, and certain B cell subsets. It mediates the directed migration and homing of immune cells by specifically recognizing and binding its ligands, CCL19 and CCL21. This interaction plays a central role in recruiting lymphocytes to secondary lymphoid organs, such as lymph nodes and the spleen, which is essential for immune surveillance, the initiation of adaptive immune responses, and the maintenance of immune homeostasis. Furthermore, aberrant expression of CCR7 is observed in various cancers and is closely associated with tumor lymph node metastasis and poor prognosis, making it a potential therapeutic target for cancer treatment and immune modulation. Its signaling pathway involves multiple downstream effector molecules, including the PI3K/Akt and MAPK pathways, which finely regulate cell survival, proliferation, and migratory behavior.
