Background
Cobolimab is a humanized monoclonal antibody targeting T-cell immunoglobulin and mucin-domain containing-3 (TIM-3). Its unique mechanism involves binding to TIM-3 to induce receptor internalization, thereby blocking interactions with ligands such as Galectin-9, reversing T-cell exhaustion, and restoring anti-tumor immunity. Clinically, the AMBER trial (Phase I/II) evaluated cobolimab as monotherapy and in combination with PD-1 inhibitors (nivolumab or dostarlimab) in advanced solid tumors. The COSTAR Lung study further explored its potential with dostarlimab and docetaxel in non-small cell lung cancer, while a Phase II trial in hepatocellular carcinoma also demonstrated promising combination efficacy. Currently, cobolimab continues to be developed as a key TIM-3 pathway candidate in immuno-oncology combination strategies.
