Background
Nectin-4 (Poliovirus Receptor Related 4) is a transmembrane protein expressed on the cell surface that primarily functions during embryonic development, with minimal expression in healthy adult tissues. However, it is found to be aberrantly overexpressed in various malignancies—such as urothelial carcinoma, breast cancer, and lung cancer—where it correlates with tumor proliferation, metastasis, and poor prognosis, making it a highly promising therapeutic target. Targeted therapies developed against Nectin-4, particularly antibody-drug conjugates (ADCs), can precisely recognize and bind to tumor cells to deliver cytotoxic payloads, demonstrating significant clinical efficacy in treating urothelial carcinoma
