Flow cytometric analysis of 2C6 expression on CAR (2C6 scFv) transfected 293F (human embryonic kidney epithelial cell) (right panel) or 293F (left panel) was stained with anti-human 2C6 scFv (Anti-CD20 CAR-T) Recombinant Rabbit mAb (FITC Conjugate) at 2 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
This antibody is a critical detection tool. Its core function is to specifically recognize and bind the 2C6 single-chain variable fragment (scFv) displayed on the surface of Anti-CD20 CAR-T cells. This 2C6 scFv is the structural domain on the CAR-T cell responsible for targeting and binding the CD20 antigen, and this antibody serves as its anti-idiotypic antibody, precisely binding to this engineered CAR molecule with high specificity, much like a "key fitting a lock." Therefore, it is commonly used as a companion diagnostic reagent in CAR-T therapy clinical trials to detect the survival, proliferation, and expression levels of infused CAR-T cells in patients via methods such as flow cytometry.
