Background

This antibody is a critical detection tool. Its core function is to specifically recognize and bind the 2C6 single-chain variable fragment (scFv) displayed on the surface of Anti-CD20 CAR-T cells. This 2C6 scFv is the structural domain on the CAR-T cell responsible for targeting and binding the CD20 antigen, and this antibody serves as its anti-idiotypic antibody, precisely binding to this engineered CAR molecule with high specificity, much like a "key fitting a lock." Therefore, it is commonly used as a companion diagnostic reagent in CAR-T therapy clinical trials to detect the survival, proliferation, and expression levels of infused CAR-T cells in patients via methods such as flow cytometry.

Flow cytometric analysis of 2C6 expression on CAR (2C6 scFv) transfected 293F (human embryonic kidney epithelial cell) (right panel) or 293F (left panel) was stained with anti-human 2C6 scFv (Anti-CD20 CAR-T) Recombinant Rabbit mAb (Alexa Fluor® 488 Conjugate) at 2 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
