Background
Adalimumab is a fully humanized IgG1 monoclonal antibody produced by recombinant DNA technology in Chinese hamster ovary cells. It neutralizes tumor necrosis factor-alpha (TNF-α) with high affinity and specificity by blocking its binding to the TNFR1 and TNFR2 receptors, thereby effectively inhibiting TNF-α-mediated pro-inflammatory signaling pathways. As an important immunomodulatory biologic agent, adalimumab is widely used in clinical practice for the treatment of various autoimmune diseases, including rheumatoid arthritis, ankylosing spondylitis, psoriatic arthritis, Crohn's disease, ulcerative colitis, and non-infectious uveitis. It is administered via subcutaneous injection, typically once every two weeks. Despite its remarkable therapeutic efficacy, the use of adalimumab requires vigilance regarding serious infections (such as tuberculosis reactivation), hepatic impairment, injection site reactions, and rare risks of malignancy; therefore, rigorous screening and assessment should be conducted prior to treatment. Adalimumab was the first fully human monoclonal antibody approved globally and is the generic name for the "blockbuster drug" Humira. Following its patent expiration, multiple biosimilars have become available, significantly improving the accessibility of autoimmune disease treatments.
