Background
AMPKβ1/2 are indispensable scaffolding isoforms of the 5′-AMP-activated protein kinase (AMPK) complex that harbor a conserved carbohydrate-binding module (CBM) which docks glycogen and related oligosaccharides to allosterically sensitize the holoenzyme to cellular energy stress, while simultaneously anchoring the α-catalytic and γ-regulatory subunits through their C-terminal sequences to ensure correct heterotrimeric assembly, stable lysosomal or mitochondrial localization, and optimal phosphorylation-mediated activation by upstream kinases such as LKB1 and CaMKK2, thereby governing systemic metabolic fluxes including fatty-acid oxidation, glucose uptake, and autophagy in liver, skeletal muscle, and adipose tissue.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-AMPKβ1/2 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
