Background
Alix, also known as ALG-2-interacting protein X or PDCD6IP, is a crucial multifunctional adaptor protein belonging to the Bro1 domain-containing family that plays a pivotal role in regulating endosomal sorting and membrane trafficking processes, particularly within the Endosomal Sorting Complex Required for Transport (ESCRT) pathway. Structurally characterized by an N-terminal Bro1 domain, a central proline-rich region, and a C-terminal V domain, Alix serves as a molecular bridge that interacts with various ESCRT-III components, such as CHMP4, and ubiquitinated cargo proteins to facilitate the formation of intraluminal vesicles in multivesicular bodies, thereby directing cellular proteins toward lysosomal degradation or exosomal release. Beyond its canonical function in membrane remodeling and cytokinesis, Alix is extensively involved in apoptotic signaling through its interaction with apoptosis-linked gene 2 (ALG-2) and participates in viral budding mechanisms, making it a significant subject of study in understanding cellular homeostasis, neurodegenerative diseases, and cancer progression.
