Flow cytometric analysis of human Granzyme B expression in Human PBMC. Human PBMC were fixed with 4% PFA and permeabilized with 0.1% Tween. The cells were stained with Phycoerythrin Mouse Anti-Human CD8 Antibody and either Alexa Fluor® 700 Mouse IgG1, κ Isotype Control (left panel) or Alexa Fluor® 700 Mouse Anti-Human Granzyme B Antibody (right panel) at 5 μl/test Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
Granzyme B is a 32-kDa serine protease stored in the cytolytic granules of cytotoxic T lymphocytes and natural killer cells, whose immune function centers on perforin-assisted translocation into target cells where it directly cleaves and activates pro-apoptotic substrates such as procaspase-3, Bid and PARP, thereby igniting both caspase-dependent and caspase-independent mitochondrial apoptosis pathways to eliminate virus-infected or malignant cells.
