Flow cytometric analysis of Human CD123 expression on human PBMC (human peripheral blood mononuclear cell). Human PBMC were stained with Alexa Fluor® 488 Mouse Anti-Human HLA/DR antibody and either Alexa Fluor® 700 Mouse IgG2a, κ Isotype Control (Left panel) or SDT Alexa Fluor® 700 Mouse Anti-Human CD123 Antibody (Right panel) at 1.25 μl/test. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
CD123, also known as interleukin-3 receptor alpha chain (IL-3Rα), is a 75 kDa type-I transmembrane glycoprotein that pairs with the common β-subunit (CD131) to form the high-affinity IL-3 receptor, triggering JAK/STAT, PI3K/AKT and MAPK cascades that drive proliferation, survival and differentiation of hematopoietic stem/progenitor cells, plasmacytoid dendritic cells, basophils and eosinophils; it is constitutively over-expressed on blasts and leukemic stem cells in >90 % of acute myeloid leukemias, B-ALL and blastic plasmacytoid dendritic cell neoplasm, making it a key diagnostic biomarker and a validated target for monoclonal antibodies, antibody-drug conjugates, bispecific T-cell engagers and CAR-T/CAR-NK therapies.
