Flow cytometric analysis of Mouse CD144 expression on bEnd.3 cells. Cells from the bEnd.3 (Mouse brain endothelioma, Right) or NIH/3T3 (Mouse embryonic fibroblast, Left) was stained with Alexa Fluor® 647 Rat Anti-Mouse CD144 Antibody Rat IgG1 κ Isotype Control (Black line histogram) and SDT Alexa Fluor® 647 Rat Anti-Mouse CD144 Antibody (Red line histogram) at 1.25 μl/test. ow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD144, also known as VE-cadherin or cadherin-5, is a calcium-dependent cell–cell adhesion glycoprotein composed of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail, expressed almost exclusively at the adherens junctions of endothelial cells in blood and lymphatic vessels, where it mediates homophilic adhesion, controls endothelial permeability, maintains vascular integrity, promotes endothelial quiescence and tubulogenesis, and is indispensable for embryonic vascular development since its genetic deletion or truncation causes mid-gestational lethality due to vessel disassembly and hemorrhage.
