Background

CD47 is a ubiquitously expressed transmembrane glycoprotein that serves as a critical dont-eat-me signal by engaging signal-regulatory protein α (SIRPα) on macrophages and dendritic cells, thereby inhibiting phagocytosis and bridging innate and adaptive immunity; beyond immune evasion, CD47 modulates integrin activation, cell adhesion, migration, apoptosis, and metabolic signaling, and its marked over-expression on diverse tumor cellsdriven by oncogenic pathways such as KRAS-PI3K-STAT3, hypoxia-HIF-1α, or amino-acid-mTOR cascadescorrelates with reduced patient survival, making blockade of the CD47SIRPα axis a promising therapeutic strategy to restore anti-tumor immunity.

Flow cytometric analysis of rat CD47 expression on SD rat splenocytes. SD rat splenocytes were stained with PE Mouse Anti-Rat CD3 Antibody and either Alexa Fluor® 647 Mouse IgG1, k Isotype Control (left panel) or SDT Alexa Fluor® 647 Mouse Anti-Rat CD47 Antibody (right panel) at 2 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
