Flow cytometric analysis of Mouse CD80 expression on LPS treated BALB/c mouse splenocytes. BALB/c mouse splenocytes treated with LPS (10μg/ml, 4h) were stained with Alexa Fluor® 647 Rat Anti-Mouse CD45R/B220 and either Alexa Fluor® 488 Rat IgG1, κ Isotype Control (Left panel) or SDT Alexa Fluor® 488 Rat Anti-Mouse CD83 Antibody at 1.25 μl/test (Right panel). Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD83 is a 45-kDa, heavily glycosylated type I transmembrane glycoprotein of the immunoglobulin superfamily that functions as a key immune modulator: its membrane-bound form (mCD83) is a cardinal surface marker of fully mature dendritic cells and is also expressed on activated B and T lymphocytes, thymic epithelial cells, neutrophils and regulatory T cells, where it is essential for efficient antigen presentation, naïve T-helper-cell priming, thymic CD4⁺ T-cell selection, and the differentiation and stability of Foxp3⁺ Tregs, whereas the naturally shed soluble form (sCD83) acts as an immunosuppressive mediator that dampens DC maturation, drives macrophages toward an anti-inflammatory, alternative-activation phenotype via the LXR pathway, and has therapeutic potential in autoimmunity and transplantation.
